It's recommended to completely discharge your battery at least once a month, preferably twice. This is not required and will not negatively affect battery performance. This is needed for phone to correctly measure remaining battery charge.
WiFi will reduce battery use, when its connected to internet. It requires less power to use WiFi, than it does 3G. If WiFi is not connected or connected to network without internet access, its better to keep it off.
