By default, does iOS Dropbox download over 3G or just Wi-Fi?

Well, um, if you are connected to WiFi, ALL your iDevices apps use said WiFi connection.
If there is no WiFi, then 3G/4G is the default...
Or did I misundersstand your question?

Well I was in the middle of downloading a huge file (750MB) and had to pop out, and wondered - if I took my phone outside, would it keep downloading over 3G. I've only got 500MB a month, ha!

I currently don't have dropbox, but I believe you may be able to restrict the apps access to mobile data and limit it to WiFi only. Have a look in settings.

I think Dropbox will drop download altogether and you will have to start over.
