Reading about the HTC One Two having on-screen buttons? Is that a good thing? Does it make much difference to Android functionality?

Nothing wrong with that per se - in fact, it means fewer hardware parts to break.
Thing is that sometimes onscreen buttons aren't as responsive as they oughta be, or too responsive. Also, obviously, they take up screen space. In some apps you'll have a row of onscreen buttons, and another row of apps specific menu buttons etc... essentially taking up 20% of screen estate...
It's a balancing act, of course.
